ABSTRACT

OBJECTIVE@#To investigate the status of anxiety and depression in patients requiring emergency treatment during the epidemic of COVID-19 to identify the patients with acute psychological stress disorder.@*METHODS@#During the COVID-19 epidemic, the medical staff divided the patients visiting the emergency department into suspected group, fever group and control group through interview of the patients at triage. Self-rating anxiety scale (SAS) and self-rating depression scale (SDS) were distributed to each patient, and a trained medical staff was responsible for assisting the patient to complete the scales.@*RESULTS@#A total of 557 sets of scales were distributed, including 211 in suspected COVID-19 case group, 167 in fever group and 179 in the control group. A total of 516 scales were retrieved, including 197 in suspected case group, 151 in fever group and 168 in control group. In the 3 groups, the incidence rates of anxiety and depression were 57.87% and 58.88%, 48.34% and 43.71%, and 18.31% and 18.99%, respectively, and the rates were significantly higher in suspected group and fever group than in the control group ( < 0.01), and significantly higher in suspected group than in fever group ( < 0.05). The standardized anxiety and depression scale scores in suspected case group, fever group and control group were 57.38±16.25 and 42.58±14.27, 51.23±15.29 and 38.32±15.39, and 32.58±17.8 and 12.25±12.94, respectively. Compared with the control group, both suspected case group and fever group had significantly higher standard scores for anxiety and depression ( < 0.01), and suspected case group had significantly higher standardized scores than fever group ( < 0.01).@*CONCLUSIONS@#Among the patients visiting the emergency treatment, the patients with suspected COVID-19 and common fever are more likely to develop anxiety and depressive symptoms.

ABSTRACT

Objective To investigate the effects of edaravone on sympathetic remodeling and ventricular arrhythmias in rats myocardial infarction model. Methods A total of 65 rats were randomly divided into the sham operation group (n=15), model control group(n=25),and edaravone group(n=25).Myocardial infarction model was established by ligation of the left ante-rior descending coronary artery. The edaravone group was injected intraperitoneally with edaravone (3 mg·kg-1·d-1), while the model control group and sham groups were injected with 0.9% sodium chloride soution.7 days after operation, the hemodynamics were detected and then ventricular arrhythmia at the peri-infarct zones was induced. After these studies, the expression of the signa-ling pathway NF-κB/p65, IκBα, p-IкBα at the peri-infarct zones were examined by western blotting, while the expression of GAP43 and TH were examined by immunohistochemical method. Results Compared with the sham group, the inducibility of ven-tricular arrhythmia after MI was increased, the expression of GAP43 and TH were increased significantly(P<0.05),the signaling pathway of NF-κB was activated, while compared with the model control group, all of the above were suppressed by edaravone(P<0.05). Conclusion Edaravone inhibits sympathetic remodeling and ventricular arrhythmias after myocardial infarction, the effects of which may be partly related to the inhibition of NF-κB pathway,(P<0.05).

ABSTRACT

Objective: To construct effective short hairpin RNA (shRNA) recombinant plasmids targeting humanDystrophin Dp71 gene, and evaluate their interference effciency. Methods: hTree pairs of siRNA sequences targeting human Dp71 gene and one pair of control siRNA sequence were designed, synthesized, and then inserted into the pRNAT-U6.1/Neo vector. hTe shRNA recombinant vectors were evaluated by enzyme digestion and sequencing. Dp71-shRNA and control shRNA plasmids were transfected into human normal gastric epithelial cells (GES-1) and humanbronchial epithelium (HBE). Western blot was used to evaluate its interfering effciency. Results: Restriction enzyme digestion and sequencing showed that the Dp71-shRNA vectors were successfully constructed. Western blot displayed that Dp71 protein expression was reduced to a signiifcant degree atfer transfection with the 3 Dp71-shRNA plasmids, and Dp71-shRNA2 plasmid inhibit the Dp71 expression most effciently. Conclusion: Dp71-shRNA vectors have been successfully constructed. The 3 Dp71-shRNA plasmids can inhibit Dp71 expression in GES-1 and HBEC, with Dp71-shRNA2 plasmid displaying the highest inhibition effciency.

ABSTRACT

Objective:To explore the clinical characteristics and diagnosis and treatment of occult poisoning caused by long-acting anticoagulant rodenticides.Method:Records of 12 patients from July 2008 to April 2011 diagnosed as anticoagulant rodenticide occult poisoning, who had been misdiagnosed initially at other hospitals were analyzed retrospectively.Elements from the records included clinical symptoms and signs, laboratory findings for prothrombin time (PT) and activated partial prothrombin time (APTT),and initial misdiagnosis and treatment outcome at our hospital. Results:The clinical presentations of patients were insidious and serious, often presented as skin ecchymose, hematuria, menorrhagia and gastrointestine bleeding.Laboratory examinations showed prolonged PT and APTT; bleeding was controlled effectively by administoring vitamin K1 daily. There were statistical difference between PT and APTT before and after the treatment (P<0.01).Conclusion:Coagulation disorders might be caused by the reduced acquisition in vitamin K dependent coagulation factors, which tends to be neglected due to a hidden medical history, delayed signs of poisoning, and various organs involved.A detailed patient history and systematic review may improve the diagnostic accuracy.Once diagnosed is made, vitamin K1 should be given as soon as possible.

ABSTRACT

AIM:To study the effect of adventitia removal on vascular structure and function in spontaneously hypertensive rats(SHR).METHODS: Thirteen-week-old male SHR(n=8) were removed their right carotid artery adventitia(SHR control group),8 age-matched male Wistar-Kyoto rats were selected as normal control group(WKY group). With all rats, the right carotid artery adventitia was removed by mechanical and chemical means, and the left carotid artery adventitia was conducted with fake operation for control. After 4 weeks, the velocity of carotid artery was detected by electromagnetic rheometer.Plasma and carotid artery angiotensin Ⅱ(AngⅡ) levels were measured by radioimmunoassay.Lumen cross section area(LA),intraelastic layer area(IELA) and extraelastic layer area(EELA) were measured by computed video processing. The ratio of(IELA-LA)/(EELA-IELA) was calculated. The expression of angiotensin converting enzyme(ACE2),extracellular signal-regulated kinase 1/2(ERK1/2) and protein kinase C-?(PKC-?) protein were determined by immunohistochemistry method. The expression of ACE2 mRNA was evaluated by reverse transcription-polymerase chain reaction(RT-PCR).RESULTS:(1) The hyperplasia of intima in adventitia removal was more significantly exacerbated than that in adventitia integrity(P
